【摘要】 目的 通过观察姜黄素对哮喘大鼠肺泡灌洗液总细胞数及嗜酸性粒细胞(Eos)计数、肺组织粒细胞-巨噬细胞集落刺激因子(GM-CSF)表达的影响,探讨姜黄素对哮喘的治疗作用。方法 36只雄性SD大鼠随机分为正常对照组(A组)、哮喘模型组(B组)、姜黄素治疗组(C组)各12只,采用卵蛋白(OVA)致敏大鼠哮喘模型,观察三组动物哮喘发作症状、肺泡灌洗液总细胞数及嗜酸性粒细胞计数情况及原位杂交方法检测肺组织中GM-CSFmRNA含量变化。结果 B组大鼠哮喘发作症状最明显,C组大鼠症状轻,A组无症状。B组肺泡灌洗液总细胞数及嗜酸性粒细胞计数均明显高于其余两组; C组明显少于B组; A组最少。肺组织中GM-CSFmRNA表达A组微弱表达, C组较强, B组表达最强。结论 姜黄素可抑制哮喘大鼠的气道炎症,其作用机制可能是通过抑制GM-CSF的表达而导致嗜酸性粒细胞生成减少及存活时间缩短来实现。

【Abstract】 Objective To observe the effects of curcumine on granulocyte macrophage colony stimulating factor expression and airway inflammation in asthmatic rats. Methods 36 male Sprague-Dawley rats were randomly divided into normal control(A group), model group(B group),curcumine group(C group), 12 rats in each group. Rats were sensitized and challenged continually with ovalbumin(OVA) for the purpose of induction of asthma in rats. To observe asthmatic attack and airway inflammation in asthmatic rats. Granulocyte macrophage colony stimulating factor in rat pulmonary tissues was detected by using in situ hybridization method. Results Asthmatic attack obviously in B group,gently in C group, Absence of symptoms in A group. The total cell amount in bronchoalveolar ravage fluid and eosin Phil were obviously higher in B group than C group and A group. The expression of GM-CSFmRNA in C group was significantly higher than that in A group and it was lower than that in B group.Conclusions The effect of curcumine on airway inflammation in asthmatic rats may be correlated with its inhibition on over expression of GM-CSF in lung tissue.医.学全.在.线网站www.med126.com

支气管哮喘是一种特殊的气道炎症,主要表现为气道高反应与变异性较大的可逆性气流阻塞。有研究表明[1],粒细胞-巨噬细胞集落刺激因子(GM-CSF)在炎症过程中起核心作用。姜黄素因其在抗炎、抗纤维化、抗肿瘤、抗氧化等作用而倍受学者们的关注[2]。本研究通过观察姜黄素对哮喘大鼠气道炎症、肺组织粒细胞-巨噬细胞集落刺激因子(GM-CSF)表达的影响,探讨姜黄素对哮喘的治疗作用。
